Abstract

In this study, we studied the effect of finasteride (5-hydroxydopamine) on the growth and viability of human prostate cancer tissue, by using a cell-specific prostate cancer cell line ( DU145). We also determined the effect of finasteride on the proliferation of prostate cancer cells. We found that finasteride did not affect the viability of prostate cancer cells. The data showed that finasteride does not affect the growth of prostate cancer cells. However, finasteride did significantly increase the viability of prostate cancer cells, and increased the number of viable cells. This suggests that finasteride was able to reduce prostate cancer cell growth by reducing the viability of prostate cancer cells. Our study showed that finasteride can affect the growth of prostate cancer cells, which may be attributed to the enhanced levels of the cell cycle regulators, such as p53 and G2/M.

Introduction

The development of novel drug delivery systems (CDSs) is a key step in the treatment of prostate cancer. The production of CDSs depends on the drug concentration and the presence of drugs, and the drug concentration can be controlled by various factors, such as the time of drug administration, the time for the drug to reach the target site, and the dose of the drug, as well as other factors. The development of CDSs for prostate cancer treatment is also a matter of concern because of the increasing evidence showing that the cancer cells can be killed by the CDSs in the presence of the drug. It is well known that cancer cells are resistant to the CDSs, and therefore the treatment of prostate cancer with the CDSs is not a viable option.

It has been reported that the effect of finasteride on prostate cancer cell proliferation is not affected by finasteride treatment. Finasteride has been reported to inhibit the growth of prostate cancer cells by reducing the levels of the cell cycle inhibitors p53 and p19. However, it has not been shown to affect the growth of prostate cancer cells by increasing the number of cells. The main aim of this study was to examine the effect of finasteride on the viability of prostate cancer cells. The study was conducted by using DU145 (human prostate cancer cell line) and PSA (prostate-specific antigen) cell lines. The DU145 cells were cultured in high-glucose Dulbecco’s modified Eagle’s medium (DMEM), supplemented with 5% fetal bovine serum (FBS), 100 U/ml penicillin and streptomycin (Gibco, Grand Island, NY, USA), and finasteride (5-hydroxydopamine) was purchased from Sigma-Aldrich (St. Louis, MO, USA). The cells were cultured in RPMI 1640 medium (HyClone, Walkersville, MD, USA), which was supplemented with 10% FBS, 100 U/ml penicillin and streptomycin.
